Analysis of mutations in the Fc region of monoclonal antibodies

Monoclonal antibodies (mAbs) are widely used as therapeutics for an increasing number of diseases such as autoimmune and any types of cancer by recognizing cell-surface proteins and killing the targeted cells by multiple mechanisms. The fragment crystallizable (Fc) domain interacts with Fc gamma receptors to activate or inhibit effector functions. In this way, engineered Fc variants were developed to adjust the Fc functions. IMGT® provides IMGT/2Dstrucutre-DB and IMGT/DomainGapAlign tool to analyse these Fc variants for mAbs published by INN/WHO. In this project, the student will develop a program to identify the mutations in the Fc region on mAb sequences present in IMGT/2Dstructure-DB. A search should be made for scientific articles that describe the main mutations found through text mining. The mutation search should be performed in scientific articles that explain the reason for the utilization for each mutation found and correlate it with the effector on binding and the modifications of effector properties of the monoclonal antibody.
