Current data indicate that condensin shapes mitotic chromosomes by translocating DNA through its ring structure, thereby extruding DNA-loops of increasing size. What remains unknown, however, is how such a DNA-loop extrusion reaction could take place in vivo, in the crowded context of chromatin. Inevitably, condensin must, in the course of loop extrusion, encounter DNA bound factors that may impact on its residence time and/or its activity, but very few condensin binding partners have been identified so far. To better understand the integrated functioning of condensin we want to search in an unbiased manner for its binding partners. With that aim in mind, we want to combine our experimental approach with an in silico pull-down approach using AlphaFold to discover new protein-protein interactions. The Master intern will systematically run Artificial Intelligence programs to discover undetected interactions between the components of condensin and the whole yeast proteome. The Master intern should have a keen interest in bioinformatics and programming in python. Also important is the knowledge of Unix systems.
